Assessment of seizure-liability using hiPSC-derived central nervous neurons in combination with the Maestro™ MEA system
A pilot study was performed to assess the effects of seizure-related compounds on the electrophysiology of Ncyte CNS neurons using the Maestro™ MEA system.
The Ncyte CNS neurons responded to the compounds tested by specific changes of their spike activity and spike pattern as expected. Below, an example is shown of the effect of the reference compound, picrotoxin, in Ncyte CNS neurons.
Picrotoxin improved the structure level in the spontaneous activity of Ncyte CNS neurons
MEA technology was used to assess effects of seizure-inducing compound picrotoxin. Raster plots illustrate the distribution of action potential detected in 9 electrodes per well over time. Picrotoxin did significantly improve the network burst coordination in Ncyte CNS neurons.
